The Particle Size Distribution in Leukocyte-reduced Red Blood Cell Suspensions in Different Durations of  Sample Storage

Abstract:  Objective  As storage time  extension, fragments and particles changes need to be explored for breakage and hemolysis of aleukocytic red blood cell suspensions, so as to provide a  experimental basis  for the transfusion safety. Methods  Aleukocytic suspending red blood cells from stocks were prepared in different storage  days (3d, 7d, 14d and 21d).  Particles in the samples according to the grain size distribution were analyzed through dynamic light scattering. Results Particles with the size  from 10 to25 nm in the supernatant of  the leukocyte-free red blood cells on 3d and 7d, while particles of 150~300 nm in size were predominantly noted after 3 weeks storage. Conclusion  Particle size measurements in the RBC suspension of leukocyte reduced storage samples would be useful for evaluation the hemolysis in RBC transfusion.
