RhD抗原量子点荧光检测研究

Develop a Quantum Dots Fluorescent Detection of RhD Blood Group

摘要: 目的 建立一种新型的RhD抗原检测方法。方法 采用量子点-抗体复合物和C1q-磁珠分离RhD抗原,根据反应前后的荧光强度比值 (Ratio value,R值) 对RhD抗原进行检测;对160例临床标本的RhD血型分型,确定量子点荧光检测法的检测性能。结果 当红细胞浓度处于1×1010~1×1011/L,35 min内可完成RhD血型抗原的检测;检测体系的组内和组间变异系数分别为3.86%和5.06%。通过量子点荧光检测法鉴定160例临床标本的RhD血型,与试管法相比符合率为100%;ROC曲线分析显示,当Cut-off值为1.610时,量子点荧光检测法在临床样本中具有较高的敏感性和特异性。结论 量子点荧光RhD血型检测可作为常规RhD抗原检测方法的一种有效补充。

Abstract: Objective To develop a new method for RhD blood group typing. Methods In the quantum dots fluorescence detection, blood group D antigen was seperated by quantum dots-antibody complexes and magnetic beads, and identified according to the ratio of fluorescence intensity before and after the reaction (Ratio value). The detection performance of this method was validated by blood typing of 160 clinical specimens. Results When the RBC concentration is at 1×1010~1×1011/L the detection of RhD blood group antigen could be completed within 35 minutes. The intra- and inter-group variation coefficients of the detection system were 3.86% and 5.06%, respectively. Compared with the tube test method, the accuracy of the quantum dots fluorescence detection was 100% by the RhD blood group results of 160 clinical samples. ROC curve analysis showed that when the Cut-off value was 1.610, the quantum dot fluorescence detection method had the highest sensitivity and specificity. Conclusion Quantum dot fluorescent RhD blood group detection can be used as an effective supplement to conventional RhD antigen detection methods.
