糖蛋白质组学定量方法用于宫颈癌血清标志物筛查的探索

A Mass Spectrometry Based Glycol-proteomic Quantitative Technique for Screening of Serum Markers in Cervical Cancer

摘要: 目的 将iTRAQ联合LC-MS/MS技术的糖蛋白质组学定量方法用于宫颈癌及正常人血清的分析,寻找差异糖蛋白,筛选潜在的宫颈癌血清标志物。方法 收集10例健康体检者和10例宫颈癌患者血浆,每组等量混合后去取部分酶解,进行iTRAQ标记,再用混合凝集素对糖肽进行富集,最后经LC-MS/MS分析。结果 共鉴定出157个糖蛋白,其中有18个蛋白存在差异表达,与正常组相比较,颈癌组血清中10个蛋白表达量上调,8个蛋白表达量下调。结论 筛选出多个与宫颈癌相关的差异蛋白,为宫颈癌血清标志物的发现提供了线索。

Abstract: Objective We used the quantitative method which combines iTRAQ with LC-MS/MS glyco-proteomics technology in the analysis for serum marker screening in cervical cancer. Methods Plasma of 10 healthy volunteers and 10 cases of cervical cancer patients were collected for mixed partial enzymatic digestion in each group, and iTRAQ labelling was performed and glycopeptides were enriched. All of the samples were subjected to LC-MS/MS analyses. Results A total of 157 glycoproteins were identified,of which 18 proteins show expression difference compared with the normal control.
