CD4+CD25+FOXP3+ Regulatory T cells in Asthmatic Mice

Objective To observe the change of CD4+CD25+FOXP3+ regulatory T cells (Tregs) in mouse model of asthma.Methods A total of 20 Balb/c female SPF-class mice were randomly divided into normal control group (n=10) and asthma model group (n=10).The asthma model group was sensitized with 2% ovalbumin (OVA) by a combination of intraperitoneal injection to induce asthma.Mice of normal control group received normal salineinstead of OVAin equal volume.The counts of eosinophils in peripheral blood (PB)and bronchoalveolar lavage fluid (BALF) weredetected.The number of Tregs in mouse spleens was analyzed.Results The numbers of CD4+CD25+/CD4+(%),CD4+FOXP3+/CD4+(%),and CD4+CD25+FOXP3+/CD4+(%) in mouse spleens in asthma and normal control groups were 2.50±0.12 vs.16.64±2.11,3.92±1.23 vs.27.72±0.15,and 1.34±0.25 vs.15.60±0.62,respectively,showing significant differences (P<0.05).Conclusion The proportion of CD4+CD25+FOXP3+ cells in asthma mice was decreased compared to controls,suggesting that tregsmay be involved in asthma pathogenesis.

Trichostatin A Combined with All-trans Retinoic Acid Induce Apoptosis in Human Ovarian Cancer Cell Line SKOV-3 SHENG

Objective To investigate the apoptotic effect on human ovarian cancer cells by both Trichostatin A(TSA)and all-trans retinoic acid(ATRA)and synergistic effect. Methods SKOV-3 cells were treated by different group of TSA and ATRA. The inhibitory rate of proliferation and the apoptosis rate were examined. Western blot was introduced to evaluate the protein expression levels. Results The proliferation of SKOV-3 cells was significantly suppressed by TSA combined with ATRA and the difference was significant compared with other groups (P<0.05). The apoptosis rate of combination group was higher than that of other groups and the difference was significant (P<0.05). Conclusion TSA combination with ATRA induces cytotoxicity in ovarian cancer cells in vitro,and the effect was much more significant than other groups.

The Dynamic Change of Various Biochemical Markers during the Preservation of Apheresis Platelets

Objective To Analyze and discuss the changes in number,morphology,and function in the preservation period of apheresis platelets.Methods Countand observe the morphologyof platelets while determinethe glucose,lactic acid concentration,platelet P-selectin (CD62p) and platelet aggregation function.Results During in vitro preservation,platelets count and PH value were significantly lower when it comes toDay 5.Platelet morphology score significantly declined since the third day.The glucose concentration gradually decreased and lactic acid concentration showed a trend of rising.Compared with Day1,platelet aggregation function gradually reduced (P<0.05) in Day 3 and Day 5.Conclusion During in vitro preservation period of apheresis platelets,the platelet morphology and function dynamically changed,but were stable in the first three days.

Objective To provide a basis for further study on platelet transfusion in patients with liver diseases. The status of platelet application was investigated in the liver special hospital. Methods Clinical and platelets transfusion data were collected for all patients who were transfused with aphaeresis platelet from Jan to Mar,2015.The indications,dosage,and effect of platelet transfusion were statistically analyzed. Results The primary diseases in platelets transfusion were liver cirrhosis and liver cancer in the hospital. Prophylactic transfusion was the main reason for platelets use,which accounted for 86.5 percent. 30.75% of platelets transfusion were performed for improving blood coagulation before intervention.Unexpectedly,infusions for treatment only accounted for 20.31%. The threshold of platelet transfusion mostly was (10~40)×109/L. The effect of platelet transfusion was not associated with cirrhosis progress. Conclusion It's suggested that the indication and dosage of prophylactic platelets transfusion should be optimized in the patients with hepatic diseases.

A Comparison of Three Blood Coagulation Assays in Patients with Component Transfusion

Objective To compare the difference of three blood coagulation function assays in patients with blood component transfusion. Methods Sixty patients who had received platelets and/or plasma transfusions were selected to carry ouy routine haemostatic assays,thrombelastogram(TEG),and Sonoclot analysis. The correlation and consistency of the three Methods were evaluated. Results A significant correlation of the R,ACT,PT,APTT and INR were observed in the three Methods. The ANGLE,CR and FIB,that reflect fibrinogen founctions,were also highly correlated,and similar correlation was noted in the MA,TP and PLT that indicate platelet founctions. Consistency of the parameters of R,ACT,and ANGLE and CR exists but the poor uniformity of the other parameters was seen. A significant correlation of TP and INR was observed with bleeding of patients. Conclusions A satisfactory correlation in routine coagulation tests,TEG and Sonoclot exists. Evaluation of coagulation founction should be based on the laboratory profile tests together with clinical menifestations of the patients.

Case Reports of N-ABO-Hemolytic Disease in Newborns Caused by Maternal Irregular Antibodies

Objective To report 10 cases of N-ABO-hemolytic disease in newborns (N-ABO-HDN) caused by maternal irregular antibodies through retrospective analysis. Methods The NEO was used to detect the irregular antibodies,and the positive samples were further identified. Cord or venous blood were collected from 74 pregnant women with irregular antibodies for hemolytic disease of newborns after delivery. Results Seventy four of 23 241 cases were found to present positive irregular antibodies. Among them,10 case of the survived infants were diagnosed as N-ABO-HDN. Two cases showed anti M-HDN; 3 cases,anti D -HDN; and 5 cases,anti E (anti Ec) -HDN. Conclusion Examination of irregular antibodies in pregnant women is quite usefull in ensuring the security of both mathers and foetuses. Early detection of irregular antibodies is crucial in multiparous pregnancies,especially in those with maternal habitual abortions.

A Comparative Observation of Serum Immunoglobulin Levels Following Blood Transfusion in Patients with Hyperbilirubinemia

Objective To explore the change of serum immunoglobulin levels before and after blood transfusion of patients with hyperbilirubinemia. Methods Sixty five neonatal patients in the hospitals were selected. They were divided into normal,mild,moderate and severe groups according to ABE scoring. The changes of bilirubin level,immunoglobul of IgA,IgM,IgG,abnormal frequencies of biochemical markers and severe by-side reactions were compared after transfusion.Results After blood exchange by transfusion,levels of total bilirubin (190.67±85.46) umol/L,indirect bilirubin(204.64 ±90.57)umol/L and albumin(31.65±3.23)umol/l were significantly lower than those before transfusion (P<0.05); the IgG in mild-severe ABE groups were high compared with that in normal patients (P<0.05). The IgA and IgM antibodies in all groups were increased (P<0.05),whereas IgG was decreased (P<0.05) after transfusion. A significant difference was seen of the incidence of hypercalcemia and hypomagnesemia after treatment among the groups (P<0.05). After blood exchange treatment,there occurred the apnea (8 cases,50.0%),respiratory failure(6 cases,9.23%),cyanosis(4 cases,6.15%),acid poisoning(6 cases,9.23%),and blood pressure fluctuations (5 cases,7.69%). Conclusion The patients with hyperbilirubinemia after blood exchange via transfusion presented decreased bilirubin and IgG,and increased IgA and IgM antibodies. Transfusion,however,may cause changes of blood markers and serious adverse reactions.

A Clinical Investigation of Matched Blood Transfusion in the Patients with Stem Cell Transplants of ABO Incompatibility

Objective To study the characteristics of the "matched blood transfusion" in the patients with stem cell transplants of ABO incompatibility.Methods Twenty five casesof ABO blood group incompatibility were detected for their blood group chimericcharacteristics,including the surface antigen of red blood cells (RBCs) and serum antibody alteration. Scheme of transfusion of chimericRBCs andthe effect and adverse reactions following transfusion were evaluated.Results Twenty five patients with stem cell transplants and ABO incompatibility received matched blood transfusion based on the surface antigen of RBCs and serum antibody alteration in chimera,and the transfusion efficacyof 76.38% was obtained. A significantly increased HB levelinpre- and post- transfusions was noted although 3 times of adverse reactions such as non-hemolytic febrile occurred. Conclusion The matched red blood cell transfusion in the patients with chimeric stem cell transplants and ABO incompatibility may help elevate the HB level and ensure the transfusion safety.

Study on the Rechecked Strategy of Antibody to Hepatitis C Virus

Objective To Investigate the rechecked strategy of chemiluminescence immunoassay(CLIA) combined with enzyme linked immunosorbent assay(ELISA)in order to detect Hepatitis C virus antibodies. Methods Two hundred and one sera samples of CLIA anti-HCV S/CO﹥0.40 were collected from the patients by ELISA. and the samples over the cutoff value with at least one reagent detection were checked for HCV RNA by realtime fluorescence quantitative PCR(FQ-PCR) . Results Among the positive 201 samples,CLIA provided by Johnson,ELISA by MUREX,and ELISA by Kehua companies revealed positivities of 2.5%,1.6%,and 1.5%,respectively. There were 75 (1.3%,75/5 800)samples that had both CLIA and two ELISA reagents positive detections. FQ-PCR positive detections were noted in 51(0.9%,51/5 800)samples. Forty samples exhibited anti-HCV S/CO﹥0.40 and <0.90 in CLIA detection. Eight samples showed anti-HCVS/CO﹥0.90 and <0.99 in CLIA detection. There were 153 samples that revealed the anti-HCV S/CO≥1.0 in CLIA detection,among them,66 samples indicated S/CO 1.00-5.00,16 samples,S/CO 5.01-10.00; 5 samples,S/CO 10.01-20.00; and 66 samples,S/CO﹥20.00. Conclusion CLIA is reliable for detection of anti- HCV with S/CO﹥0.40 and <0.90. HCV RNA detection is useful for laboratory diagnosis and treatment evaluation in the samples that have showed positive anti-HCV by using CLIA and ELISA.

Distribution of NDM-1 Gene in Strains of Acinetobacter sp

Objective To investigate the distribution of NDM-1 in Acinetobacter spp. Methods Twenty-five isolates of Acinetobacter baumannii and 3 isolates of A. lwoffii and 2 isolates of A.junnii that were resistant to imipenem were selected for the study. Carbapenemases were screened by modified Hodge test and EDTA disk synergy test. The genotypes of carbapenamases were determined by PCR and DNA sequencing. Plasmid conjugation was performed to study the transfer of carbapenem resistance. Strain homology was analyzed by PFGE. Results Ten out of 25 A. baumannii strains were positive for MHT but all were negative for EDTA disk synergy test,and all the 25 strains were positive with OXA-23-like but negative for NDM-1. All the non-A.baumannii isolates were positive for EDTA disk synergy test but negative for MHT. The strains were just positive for NDM-1 detected by PCRs and negative for the other carbapenamases. The NDM-1 positive strains had different PFGE profiles. PFGE showed high homology for A.baumannii with mainly A clone. Conclusions NDM-1 was distributed in the bacterias rather than A. baumannii,which were resistant to imipenem and the positive isolates had different PFGE profiles.

Application of Scatter Diagram of Hematology Analyzer to Survey of False Increase of Basophil Count

Objective To analyze the change of the scatter plots of white blood cell and basophilic granulocyte count channels (WBC/BASO)and explore the resean for false elevation of basophils count. Methods The data of scatter diagram of false elevation in Baso counts were collected that had been verified by manual counting in blood smears of 46 impatients. Results Three factors contributed to the false increase of Baso count,including abnomal lymphocyte (Abn-L),neutrophil toxic granules (TG)and improper blood collection. Among them,Abn-L interference dominates(56.5%). All of the samples of false elevated Baso counts with typical differential luekocye counts and Abn-L or TG WBC/Baso scatter diagram presented abnormal lymphocyte count or TG by manual counting in periphera blood smears. Differential leukocyte counting (DC)showed that the abnormal increase was originated from Abn-L or TG. Conclusion The samples with false increase of Baso count should be comfirmed by instrumental typical scatter diagram to provide orientation of repeated detection and correct data for clinical use.

Analysis of Relationship between Particle or Mass Concentration of Lipoprotein(a) and Character of Atherosclerotic Plaques

Objective To compare the particle or mass concentration of lipoprotein(a) and the character of atherosclerotic plaques and to analysis the effect of lipoprotein(a) on the formation of atherosclerotic plaques. Methods A retrospective study of total 91 patients who underwent computed tomography angiography (CTA) in our hospital from January 2015 to August 2016 were performed. Based on the result of CTA,those patients were divided into two groups,the control group and the coronary atherosclerotic heart disease (CAHD) group. The particle and mass concentration of lipoprotein(a) were compared between two groups. Additionally,according to the character of atherosclerotic plaques,patients in the CAHD group were further divided into three group,comprised of the soft plaque group,the mixed plaque group and the calcific plaque group. The particle and mass concentration of lipoprotein(a) in these three groups were also compared. The correlation between particle concentration and mass concentration of lipoprotein(a) was analyzed. Results The particle and mass concentration of lipoprotein(a) were significantly higher in the CAHD group compared to the control group (t=3.4707,P=0.0008 and t=3.1524,P=0.0022). The obvious difference of particle and mass concentration of lipoprotein(a) was observed in three groups of different character of atherosclerotic plaques (F=55.175,P<0.0001 and F=6.124,P=0.004). Among those three groups,patients in the calcific plaque group had the highest level of particle and mass concentration of lipoprotein(a) (P<0.05). The correlation between the particle concentration and the mass concentration of lipoprotein(a) was seen (r=0.5646,P<0.0001). Conclusion Elevated level of lipoprotein(a) could be seen in CAHD patients,and the level of lipoprotein(a) has relationship with the character of atherosclerotic plaques. Lipoprotein(a) could play different roles in the formation of different kind of atherosclerotic plaques.

The Diagnose Value of Combining Determination Vascular Endothelial Growth Factor and β-HCG in Early Non-typical Ectopic Pregnancy

Objective To determine the level of VEGF and β-HCG in early non-typical ectopic pregnancy (EP) and investigate the combining determination of diagnostic value of VEGF andβ-HCG for EP. Methods 140 pregnancy patients from Nanjing Yuhua hospital were enrolled into this study,of which 60 patients with EP (<7-week)and 80 patients with intrauterinepregnancy. The VEGF andβ-HCG levels were tested by ELISA,ROC was used to cut-off value and all the Results were analyzed by the statistical method. Results The levels of VEGF in EP was significantly higher than that in intrauterinepregnancy,while β-HCG was significantly lower than that in intrauterinepregnancy(VEGF:270.35±111.55pg/ml vs. 125.94±42.38 pg/ml,P=0.000;β-HCG:9 790.35±1 620.79 vs. 12888.13±2 544.41U/ml,P =0.000). With a cut-off value of 202.50 pg/ml,the AUC value of VEGF is 0.945. The sensitivity of VEGF testing for diagnosing EP was 75.0%,the specificity was 97.5% and the accuracy was 90.2%.With a cut-off value of 10 950.30 U/ml,the AUC value of VEGF is 0.855. The sensitivity of β-HCG testing for diagnosing EP was 76.3%,the specificity was 76.7%,and the accuracy was 83.1%. The sensitivity of the combined testing was 91.3%,the specificity was 95.0% and the overall accuracy was 85.5%. The sensitivity and the overall accuracy of combined testing were higher than those of β-HCG and ADAM12-S testing single. Conclusion EP presents higher VEGF and lower β-HCG concentration than those with intrauterinepregnancy and determination of VEGF and β-HCG is useful to differentiate EP from intrauterinepregnancy. The combined testing of VEGF and β-HCG can increase the sensitivity of diagnosis of EP.

Analysis of Nucleic Acid Tests in HBsAg,Anti-HCV,Anti-HIV ELISA Negative or Single Reagent Reactive Blood Donors

Objective Nucleic acid tests were performed in the blood donors and compared with the Results of ELISA double negative and single reagent reactive in HBsAg,Anti-HCV and Anti-HIV so as to provide a basis for distinguishing the false positive blood donors. Methods Two ELISA reagents were used to detect seral HBsAg,HCV and HIV in the donors,and 8 specimens of ELISA negative and single reagent positive samples were subjected to DNAs detections. All DNAs positive samples were identified. Results In 31 065 blood samples,30 254 cases were found to be negative with double ELISA,and 241 cases were positive with single reagent examination,among them 180 cases were detected with imported kits and 61 with domestic reagents. A significant difference was seen in different reagents. DNAs detection showed 54 positive samples including 46 HBV,3 HCV,and 5 HIV. Identification tests revealed 39 positive HBV,among them 37 and 2 cases were found from ELISA negative and single reagent positive samples,respectively. Neither HCV nor HIV was discovered and total positive rate was 72.2%.Conclusion ELISA single reagent reactive result gives rise to a high false positivity while DNAs examination may improve the efficacy of virus diagnosis. Complementary examinations are recommended so as to induce the risk of blood transfusion of infectious diseases.

Serum Adhesion Molecules SE-Cadherin、SE-Selectin and sIntercellular Adhesion Molecule-1 as Lung Cancer Biomarkers

Objective In the present study,we investigated the role of soluble E-cadherin (SE-CAD),soluble E-selectin (SE-SEL) and soluble intercellular adhesion molecule-1 (sICAM-1) as biomarkers in lung cancer. Methods 168 patients with recently diagnosed lung cancer,98 with small cell lung cancer(SCLC),and 70 with non-small cell lung cancer (NSCLC),50 cases of patients with benign pulmonary diseases as well as 50 healthy volunteers were enrolled. Blood samples were collected at the time of diagnosis and measurement of soluble adhesion molecules in the serum samples was performed by enzyme-linked immunoassay using monoclonal antibodies against E-cadherin,E-selectin,and ICAM-1. Results Serum levels of SE-CAD,SE-SEL,and sICAM-1 in both SCLC and NSCLC were significantly higher than the healthy subjects and the group with benign pulmonary diseases (P<0.05). In addition,patients with SCLC or NSCLC with distant metastasis had a marked increase of SE-CAD(P<0.05),but no such correlation with SE-SEL and sICAM-1 was found. Conclusions Our findings suggest that SE-CAD,SE-SEL,and sICAM-1 have an adjunctive diagnostic role in lung cancer. Furthermore,SE-CAD may also have a prognostic value and could be a useful biomarker in the prediction of lung cancer outcome.

Objective To investigate the bacteriology characteristics and drug resistance of post-operative infections of wounds in the patients with colorectal cancer and to provide the basis for the reasonable application of antibiotics.Methods A retrospective analysis of the pathogenic bacteria distribution and drug resistance in112 colorectal cancer patients,who underwent surgeries in the hospital from Sep 2012 to Dec 2015,was performed. Results A total of 121 strains of pathogenic bacteria were isolated from wound secretions and abdominal puncture fluids. Gram-negative bacterias accounted for 83.47% (101/121), including Escherichia coli,Pseudomonas aeruginosa,Klebsiella pneumoniae,and Enterobacter cloacae. Gram-positive cocci accounted for 13.22% (16/121),that dominated by Enterococcus. It was found that E. coli and K. pneumoniae had a low resistance rate to carbapenems,β-lactamase inhibitor compound agents,cephamycins and fourth generation Cephalosporins (less then 30%),whereas P.aeruginosa showed higher sensitivity to antibacterial agents in vitro. Enterococcus had a high resistance rate to erythromycin and tetracycline,while no drug-resistant strains to vancomycin,linezolid,tigecycline and nitrofurantoin were found. Conclusion The most common pathogenic bacterias causing surgical site infections are gram-negative bacilli. It is necessary to energetically prevent and treat postoperative infections in colorectal cancer patients based on the Results of drug susceptibility testing.

Effect of One-stop CT on Antiplatelet Aggregation in Ischemic Stroke

Objective To explore the effect of one-stop CT on platelet aggregation in different ways of ischemic stroke. Methods Ninety six patients with acute ischemic stroke (AIS)and hyperacute ischemic stroke(HIS)were treated in the hospital from 2014 to 2016. Patients with AIS were randomly divided into groups A,B and C while those with HIS included groups D,E and F. Both groups A and D were treated with aspirin enteric-coated tablets alone at 100 mg/ day whereas groups B and E were with clopidogrel sulfate at 75 mg/day,and groups C and F were given aspirin at 100 mg/day combined with clopidogrel at 75 mg /day. The absolute value of NIHSS,mRS,and ASPECTS scores were observed pre- and post-treatments,and the absolute value of infusion imaging parameters and clinical efficacy were observed. Results The absolute value of score in groups A and B was significantly lower than that in group C,and the value of group A was significantly lower than that of group D. The absolute value of score in group B was significantly lower than that in group E(P<0.05). The absolute value of perfusion imaging parameters in groups A and B was significantly lower than that in group C,and the value in group A was significantly lower than that in group D. The infarcted imaging parameters in group B were decreased compered with those in group D whereas the value in group C was lower than that in group F(P<0.05). The total effective rate of 96.87% in group C was significantly higher than that in groups A ( 84.38%)and B (83.33%). The total effective rate in group A was 84.38%,significantly smaller than that in group D (87.50%). The effective rate in group B was 83.33%,which was much lower than that in group E (84.38%),and the effective rate in group C was 96.87%,smaller than that in group F (100.00%)(P<0.05). No significant difference was seen between groups A(84.38%)and B(83.33%)(P> 0.05). Conclusion One-stop CT before and after treatment were related to the clinical efficacy. Combined treatments may obviously decrease platelet aggregation and improve neurological function. The efficacy of the three Methods treatment in patients with hyperacute stroke was better than that of acute stroke patient.

The Application Value of Platelet VEGF and TSP-1 Detections in the Lung Cancers

Objective To investigate the clinical application of platelet vascular endothelial growth factor(VEGF)and TSP-1 determination in the progression of lung cancers. Methods The levels of VEGF and TSP-1 in platelets and platelet-poor plasma(PPP)in 71 patients with lung cancers and 35 healthy controls were determined by ELISA method. Results The levels of platelet VEGF and VEGF/TSP-1 in patients with unmetastatic lung cancers and metastatic lung cancers were significantly higher than those in healthy controls (P<0.05); the levels of platelet VEGF and VEGF/TSP-1 in patients with metastatic lung cancers were significantly higher than those in patients with unmetastatic lung cancers (P<0.05). Moreover,the levels of platelet TSP-1 in patients with unmetastatic lung cancers and metastatic lung cancers were remarkably lower than those in healthy controls (P<0.05). However,the PPP VEGF and TSP-1 in the patients and control group were undetactable (<7.18 pg/ml). Conclusions The balance between the levels of VEGF and TSP-1 is shifted in the peripheral blood of patients with lung cancers,and the levels of platelet VEGF and VEGF/TSP-1 will help to reflect the progression of lung cancers.

Objective Analysis the coagulation function of a perinatal pregnant with congenital VII deficiency and the discussion of the treatment strategy. Methods Blood samples from a pregnant with congenital factor VII deficiency at the different time points in peripartum period was collected,plasma clot formation is used to determine the prothrombin time (PT),activated partial thromboplastin time (APTT),thrombin time (TT),Clauss method is used to determine the fibrinogen (Fbg),partcle-enhance immunoturbidimeric assay is used to determine D-dimer (DD)and antithrombin- III. Results Before the prophylactic substitution treatment in peripartum period,PT was prolonged and Fbg was mild rised,the other index was normal,recombinant activated factor VII (rFVIIa)was given to patients within 1 hour in preoperative,the pregant is characterized by PT shorten and presented high coagulation state. Intraoperative coagulation function was normal and no bleeding complications; postoperation coagulation function became abnormal,characterized by PT extension,higher DD and after 3 days PT was recovered to preoperative levels. Conclusion It may be essential to deteced coagulation function in the perinatal pregnants with congenital factor VII deficiency dynamically,especially the change of PT,such this case of cesarean delivery. Other indexes of coagulation and fibrinolytic may be judge patient's coagulation function effectively.
