A Case Report of High Pressure Ozone Autologous Blood Transfusion Therapy for Hypersomnia in Patients with Brain Injury

Objective To observe the effectiveness of high pressure ozone autologous blood transfusion (HPOABT) for the treatment of hypersomnia of the patients with traumatic brain injury. Methods Drug treatments included nerve growth factor (18 μg,qd.),oxiracetam capsules (0.4 g,qd.),maleic acid peculiar to Guangxi pp injection (0.32 g,qd.),Xueshuantong injection(Chinese patent medicine,0.45 g,qd.),physical and electric therapy,exercise of hemiplegic limbs,combined with 10 times of HPOABT. Results After 2 courses of treatments,both mental and physical conditions of the patients were obviously improved. The cognitive function and limbs muscle strength were increased to Ⅳ+ following 10 times of treatments. Conclusion The therapy of drugs combined with high pressure ozone autologous blood transfusion therapy,due to its effectiveness and safety,is promising for treatment of hypersomnia of patients with brain traumatic injury.

An Impact of Allogenetic Blood Transfusion on the Serum Level of Vascular Endothelial Growth Factor

Objective To investigate the influence of blood transfusion on serum levels of vascular endothelial growth factor (VEGF). Methods One hundred and twenty patientswere detected and analyzed for VEGF levels with SPSS17.0 softwarebefore or after transfusion. Results The VEGF level of females was found to be significantly higher than that of males following blood transfusion(P<0.05). No difference was noted in age and blood group. It was fund that the VEGF expression of all subjects investigated seems to be decreased after transfusion compared with those before transfusion.(P<0.01). Conclusion The VEGF level in the sera of patients was reduced after transfusion,suggesting a possible inhibition of VEGF expression following transfused blood products.

Application of Intraoperative Blood Salvage Combined with Reinfusion of Postoperative Drainage in Lumbar Spine Surgery

Objective To investigate the application of intraoperative blood salvage combined with reinfusion of postoperative drainage in lumbar spine surgery. Methods 115 cases of lumbar spine surgery were divided into two groups:control group (73 cases) and observation group (42 cases).The control group received intraoperative blood salvage,and the observation group received intraoperative blood salvage combined with reinfusion of postoperative drainage.Hemoglobin concentration was monitored respectively 1 day before surgery,3 days and 7 days after surgery.All patients were recorded for intraoperative blood loss,intraoperative return volume,and allogeneic blood transfusion.The observation group was also recorded in postoperative drainage volume and postoperative return volume.The number of allogeneic blood transfusion during operation and postoperative was statistically analyzed. Results The amount of blood transfusion in observation group was lower than that in control group (P<0.05).There was no significant difference in blood loss,intraoperative return volume,the number of allogeneic blood transfusion,hemoglobin concentration before and after operation (P>0.05). Conclusion Intraoperative blood salvage combined with reinfusion of postoperative drainage can significantly reduce the amount of allogeneic blood transfusion,but can not reduce the number of allogeneic blood transfusion.

Experimental Study on the Change of Organism Lipid Peroxidation Level in Short Term Rapid Blood loss

Objective Study on the changes of organism lipid peroxidation level in short term rapid blood loss. Methods The rapid blood loss model of the gradient decerease in the systolic blood pressure was eatablished in 11 new zealand rabbits on ordinary level. The arterial blood during the different phase of shock,i.e.when the systolic pressures were 2/3(T2),1/2(T3),1/3(T4)of the basis systolic blood pressure,was collected and malondialdehyde(MDA)、hydrogen peroxide enzyme (CAT)、glutathione oxidation reductase (GSH-P x)levels in the plasma were determined by the double antibody sandwich method of enzyme-labeled immunosorbent assay(ELISH),and the data be statistically analyzed. Results MDA :Group B less than in group C,group C less than in group D,there was no significant difference between the time level(P>0.05). CAT :Group A higher than in group B,group B higher than in group C,group C higher than in group D,there was no significant difference between the T1-T2,T2-T3,T3-T4(P>0.05),there were significant differences between the other groups (P<0.05). GSH-Px :Group A higher than in group B,group B higher than in group C,group C higher than in group D,there was no significant difference between the T1-T2(P>0.05),there were significant differences between the other groups (P<0.05). Conclusion Short term rapid blood loss has a significant effect on the antioxidant enzyme CAT and GSH-Px of organism,can lead to significantly decrease in CAT and GSH-Px activity,MDA levels rise reflect the degree of lipid oxidation. There were significantly correlated between blood loss and CAT,GSH-Px index.

Effect of Two Kinds of Blood Preservative Solution on the Content of Factor Ⅷ Recovery in Fresh Frozen Plasma

Objective To investigate the fresh frozen plasma (FFP)VIII derived from the whole blood with CPDA or ACD-B blood preservative solution,content difference and analyze the relative factors. Methods 48 cases of FVIII derived from the whole blood with CPDA (A group) or ACD-B (B group) blood preservative solution were randomized for the determination respectively,compared the two groups of the mean value and qualified rate of FVIII. Results There was no significant difference between the 2 groups of FVIII content (P>0.05),compared with the A group,the qualified rate of FVIII quality standards in B group decreased significantly. Conclusion The decline of qualified rate of FVIII quality standards in B group related to the related factors such as the decrease of the concentration of dilution caused by the increase of the liquid storage capacity,the actual blood collection and so on.

An Observation on Blood Coagulation Indexes of Four Different Plasma

Objective To investigate the changes of coagulation indexes in different plasma after melting,and to understand the changes of plasma coagulation factors,and to provide theoretical basis for clinical use. Methods Sysmex-CA-1500 Automatic Coagulation Analyzer was used to detect test 83 plasma samples's PT、APTT、TT、 FIB at the 0.5 hours,6hours,12hours,24hours after melting. Result Removing cryoprecipitate of plasma's results are differences from the other three plasma in 0.5h . The Fib and TT results of Virus inactivation plasma and remove cryoprecipitate of plasma are different from fresh frozen plasma and nomal frozen plasma results in general 0.5 to 24 hours; almost results are no difference between fresh frozen plasma and nomal frozen plasma. Conclusion The plasma of moving cryoprecipitate can not use the same as the other three plasma. Virus inactivated plasma can only be used as a normal frozen plasma.

Epidemiological Characteristic of HIV Infection in Wuxi Volunteer Blood Donors

Objective To investigate the epidemiological features of volunteer blood donors infected with HIV in Wuxi,and to reduce the risk of blood-borne infections. Methods Data of the blood donors with HIV infection from 2011 to 2015 were collected,and the features of homosexual and non-homosexual individuals were analyzed in parallel. Results Fifty-five donors were confirmed to have HIV infection in the survey. Most HIV carriers were single males and had nonlocal residence,and gave their first donations personally or in group between the age of 20 to 40 years. Workers and farmers and those with high education dominated. Seven persons were found to have underling syphilis. In addition,the majority of the HIV infected homosexuals were local residents and workers when compared to the non-homosexuals who were farmers and nonlocal residents(P=0.042,P=0.031). Conclusion HIV infection tends to spread from high-risk persons to general population. Measures must be taken based on the characteristic of the infection to ensure the blood safety.

A Clinical Analysis of the Parameters of Erythocytes and Reticulocyte in Patients with Renal Anemia

Objective To study the changes of erythrocyte and reticulocyte parameters in patients with renal anemia. Methods Fourty three patients with anemia associated with renal disfounction(ARD) were collected for for investigation in the hospital from December 2015 to December 2016. The equal number of the patients with anemia caused by hemorrhage rather than renal disorders (AHD)were taken as control. The parameters of red blood cells (RBCs) and reticulocytes between the two groups were compared. Results The LFR of reticulocytes in ARD was significantly higher while the RET,RET#,IFR,MFR and HFR were lower than that in AHD (P<0.05). No significant difference of Hb level between the two groups was observed (P>0.05). Similarly,no remarkable differences of HCT,MCHC,RDW-SD and RDW-CV was seen between the two groups (P>0.05). The levels of MCV and MCH in ARD were notably higher than those in AHD,and the RBCs counts in ARD group was lower than that in AHD (P<0.05). Conclusion The parameters of RET,RET #,IFR,MFR and HFR of reticulocytes in patients with renal anemia are significantly decreased,while the levels of MCF and MCH in blood erythrocytes were increased. Parameters of both red blood blood cells and reticulocytes may be used as an important criterion for the diagnosis of renal anemia.

Genotyping and Drug Resistance of Biofilm Forming Strains of Klebsiella Pneumoniae

Objective To study the distribution and drug resistance ofKlebsiellapneumoniae in hospital and analyze the genotypes of K.pneumoniaewith biofilm. Methods One hundred and sixty strains of clinical specimens were collectedfrom August 2013 to August 2015.The bacterial distribution,drug resistance and K.pneumoniae genotype were analyzed. Results K.pneumoniaedominated in the specimensfrom respiratory tract and urethra,accounting for 59.38% and 22.88%,respectively.The highest rate of ICSI was 40.63%,followed by 16.25% and 14.38%.K.pneumoniaeresistance to ampicillin accounted for 80.00%,followed by cefazolin(67.50%),and ceftriaxone,(65.00%).More than 50% of the strains tested showed relatively resistant to ampicillin/sulbactam,sulfamethoxazole and cefotaxime.Antibiotics to which K.pneumoniaeproduced less resistance included erpicane,followed by imipenem and cefotetan.The biofilm positive rate in all 160 strains was 37.50%.The ESBLs detected 16 strains of K.pneumoniae biofilm,41.75% of the bacteria presented positive TEM gene and 31.25%,SHV gene while bacteria carrying both SHV and TEM genes accounted for 12.50%.No CTX2Mwas detected. Conclusion The incidence of K.pneumoniae infection in intensive care unit is relatively high,and the drug resistance of the bacteria seems to be low.The predominant genotypes of K.pneumonia with biofilm include SHVand TEM.

Objective To understand the current enzyme production and drug resistance of A.hydrophila forclinical treatment of A.hydrophila infection and to provide the basis for the rational use of antibiotics. Methods Fifty strains of A.hydrophilabacteria were collected from clinical specimens from 2011 to 2015.The routine drug sensitivity test was carried out using the K-B paper method,and the extended-spectrumβ-lactamases(ESBLs) was confirmed by the CLSI standard double disk tests.The AMPC was detected by three-dimensional test. Results Among 50 strains of A.hydrophila,15 strains produced ESBLs and 10 strains produced AMPC.All bacteria showed ampicillin resistance.Ampicillin/sulbactam resistant rate was 94%,and the third and the fourth generation cephalosporin resistant rate was 40% although the other antibiotics remained highly sensitive. Conclusion Clinical treatment of A.hydrophila infection should be careful with cephalosporins so as to improve the efficacy of antibiotics.

Application of EBV-DNA Detection to Staging of Untreated Nasopharyngeal Carcinoma

Objective To investigate the correlation between plasma EBV-DNA level and clinical stage of untreated nasopharyngeal carcinoma (UNPC). Methods Clinical stage of the UNPC patients was determined by tumor node metastasis (TNM),and the content of EBV DNA in 57 UNPC patients was detected by fluorescence quantitative PCR. Result EBV-DNA positive rate was not correlated with gender and age of patients,but plasma EBV-DNA concentration of stage Ⅲ patients was higher than stage Ⅳ patients (P<0.05). EBV-DNA positive rate of patients was increased with the development of T stage progress. The EBV-DNA concentration of stage T4 patients was higher than stage T2 patients (P<0.05). No significant difference of plasma EBV-DNA concentration between the stage M and stage N patients. Conclusion The level of plasma EBV-DNA concentration is positively correlated with clinical stages of nasopharyngeal carcinoma. Detection of plasma EBV-DNA by fluorescence quantitative PCR is a useful auxiliary method for UNPC diagnosis.

Analysis of Blood Lipid Related Indexes in Healthy People Under 35 Years Old in Suzhou

Objective To analyze the possible causes of lipid metabolism disease,we investigated the incidence of hyperlipidemia,fatty liver and hyperglycemia in healthy people under the age of 35. Methods The healthy people under the age of 35 were collected and generally analyzed according to gender the incidence of hyperlipidemia,hyperglycemia and fatty liver; and on the basis of biochemical indexes in patients with fatty liver and no-fatty liver patients were further analyzed statistically. Results The rate of TG,TC,LDL,GLU increased is 39.6%,10.33%,25.7%,3.71%,respectively,in male population,which were higher than those in female population; the detection rate of 42.38% patients with fatty liver is much higher in male population than that of female patients with fatty liver (22.41%);biochemical indexes were further analysed in patients with fatty liver and no-fatty liver,compared with the no-fatty liver group,the levelof TG,TC,LDL,GLU was significantly increased in fatty liver group,the difference was statistically significant (P<0.01). Conclusion High blood lipids,fatty liver and high blood glucose were risk factors of atherosclerosis and thrombosis,the incidence showed younger,suggesting that the health education and health examination need to further be strengthened to play a role in prevention of this disorder.

The Urine Iodine Level in 246 Midtrimester Pregnant Women in Hefei,Anhui Province

Objective To investigate the iodine nutritional status of the midtrimester pregnant women in Hefei after the new implementation of salt iodine policy. Methods Urine iodine levels were measured in 246 cases of the midtrimester pregnancies from Maternal and Child Healthcare Hospital of Hefei to analyze their iodine nutrition from December 2016 to January 2017. Results The median iodine level in urine was 143 ug/L,the ratio of iodine deficiency was 55.28%,the ratio of normal iodine level was 24.39%,and iodine level above the normal was 16.26%. 4.07 percent of the pregnant women showed an excessive iodine concentration in the urine. No statistically significant difference was seen of the urine iodine level in different age groups. Conclusion The iodine deficiency of pregnant women was notable after the implementation of the new salt iodine policy,indicating that urgent attention should be paid to optimize the daily amount of iodine uptaken to improve the quality of births.

The Value of a Combination Detection of Anti-MIT3,Anti-gp210 and Anti-sp100 in the Diagnosis of Primary Biliary Cirrhosis

Objective To Detect antibodies against mitochondrial and nuclear antigens for diagnosis of primary biliary cirrhosis (PBC) and to compare the performance of the PBC Screen to the combined performance obtained with individual IgG ELISAs to MIT3,gp210,and sp100 in the patients with PBC. Methods The PBC Screen and individual antibodies to MIT3,anti-gp210,and anti-sp100 were measured in sera of 220 individuals. Final diagnosis of PBC had been made in 95 of the participants and the rest included individuals with autoimmune hepatitis (AIH)/ primary sclerosing cholangitis (18 cases),viral hepatitis(32 cases),or other liver diseases (21 cases),and healthy controls(54 persons). Results The PBC Screen at the cutoff of 25 units had an overall sensitivity of 76.8%,specificity of 95.2%,similar to the specificity of 96.0% obtained by the combined results of individual MIT3,sp100,and gp210 IgG ELISAs. Of the 32 PBC patients without AMA detected by indirect immunofluorescence(IIF),15(46.9%) were positive for PBC-specific autoantibodies. Conclusion sThe PBC Screen is an ideal first-line test for the diagnosis of PBC,especially for those who showed negative IIF.

IGF-I and IGFPB-3 in Different Types of Brain Tumors

Objective To investigate the value of detecting the expression serum levels of insulin-like growth factor-I (IGF-I) and insulin-like growth factor binding protein-3(IGFBP-3) in patients with brain tumor. Methods Serum levels of IGF-I and IGFPB-3 were detected by chemiluminescence in several patients with brain tumor,Of which 16 cases of meningioma,13 cases of glioma,32 cases of GH secreting pituitary adenoma,40 cases of non-GH secreting pituitary tumors and 30 healthy controls. Results The serum level of IGF-I and IGFPB-3 in group of meningiomas and GH secreting pituitary adenoma were higher than that in the healthy control group (P<0.05); The serum level of IGF-I and IGFPB-3 in GH secreting pituitary tumors were higher than that in non-GH secreting pituitary adenoma; The serum level of IGF-I in GH secreting pituitary tumors preoperative were higher than that after surgery (P<0.05). Conclusion (1) The serum level of IGF-I and IGFPB-3 increased in many patients with brain tumors,detecting their level can assist the diagnosis of certain brain tumors; (2) The serum level of IGF-I and IGFPB-3 was significantly different in secreted and non-secreting pituitary tumors,which can take differential diagnosis on different type of pituitary tumor; (3) The serum level of IGF-I and IGFPB-3 in GH secreting pituitary tumors preoperative were significantly higher than that after surgery,which can monitor the effect of surgery and observe patient recovery situation by detecting the speed and extent of IGF-I decline after surgery.

Clinical Significance of PF4,VEGF,HIF-1a and Lpα in Sera of Patients with Hepatocellular Carcinoma

Objective To investigate the levels and clinical significance of platelet factor 4 (PF4),vascular endothelial growth factor(VEGF),hypoxia inducible factor-1α(HIF-1α) and lipoprotein α(Lp α) in hepatocellular carcinoma (HCC). Method The investigation included 102 inpatients of chronic hepatitis B(HCB),51 of liver cirrhosis,37 of HCC,and 186 subjects of healthy controls(HC) between 2014 and 2016. All samples were detected with immune enhanced turbidimetric method for serum concentration of Lpα and with ELISA for seral PF4,HIF - la and VEGF. Results The PF4,VEGF,HIF-1a and Lpα concentrations in HCC patients were obviously higher than those of CHB and LC patients,and HC as well (P<0.001~0.05),showing a stage-dependent increase of the markers tested(t=7.951-19.086,P<0.001-0.05). No remarkable variation was noted of PF4,VEGF,and HIF-1α in the three groups(P>0.05). Lpαlevel,however,was found to be higher in LC group than in the other groups(P<0.05),reversely showing a stage-dependent decrease(P<0.05). Conclusion sPF4,VEGF,HIF-1α and Lpαlevels are significantly elevated in the sera of HCC patients,based on which a differential diagnosis between LC and HCC can be made.

Methods A rare HLA-B allele was initially detected by Luminex PCR-SSO typing,then the sample was sequenced by sequence-based typing (SBT) and the group-specific sequencing primer(GSSP) to confirm the mutation allele and locus. Results The sequence of the novel allele was different from that of B*67:01:01 at the position 370(G>A) in exon 3,which results in an amino acid change from Gly to Ser at codon 100. Conclusion This allele is a new HLA-B allele which has been designated as HLA-B*67:07 by the World Health Organization(WHO) HLA Nomenclature Committee.

Polymorphism of Rh and Duffy Blood Group Antigens in Erythrocytes of Dai Population in Yunnan

Objective To investigate the distribution and antigen frequencies of Rh and Duffy blood group antigens in Dai nationality in Yunnan,so as to provide an evidence for the prevention and treatment of blood type-associated hemolytic diseases. Methods Two hundred cases (100 male and 100 females) of Dai people without consanguinity were collected and 100 Han people were taken as control. Blood samples (5 ml,EDTA anticoagulants) were detected for Rh,Duffy and antibody screening. Duffy blood group genotyping was performed using PCR-SSP. Results The RhD positive rate was 98.5% and the negative rate was1.5% in 200 Dai persons compared with the positive rate of 99.0% and negative rate of 1.0% in Han population. Duffy genotyping revealed the distribution of phenotypes of Fy(a + b-) (187/200,93.5%),Fy(a+b+)(12/200,6.0% ),and Fy(a-b+)(1/200,0.5% ),no Fy(a-b-) phenotype was seen,and the genetic frequencies were found to be Fya(0.965) and Fyb(0.035). In Han population,Duffy phenotypes include Fy(a+b-) (92/100,92.0% ),Fy(a+b+)(5/100,5.0% ),and Fy(a-b+)(3/100,3.0% ),no Fy(a-b-) phenotype was noted,and the genetic frequencies are Fya(0.945) and Fyb(0.055). The Dai population presented the prevalence of positive antibodies of 1.5%,among them 2 cases showed E antibodies. Duffy genotyping results showed that,in the 200 samples of Dai nationality,the gene frequency was Fya (0.965),and Fyb (0.035). The frequency of the gene was Fya (0.945) and Fyb(0.055) in 100 samples of Han nationality. Conclusion The antigen distribution of Duffy blood group system in Dai nationality is similar to those in Han population although the negative RhD and irregular antibodies are slightly higher in Dai people(P<0.05).

The Distribution of Gene Frequency of Duffy Blood Type in Uygur Population in the North Area of Xinjiang

Objectives To investigate the polymorphism of Duffy blood type in Uygur population of Northern Xinjiang so as to establish the database of rare blood type antigens of Uighur population for prevention of hemolytic diseases of newborns. Methods The Fya and Fyb antigens were found in 894 randomly selected Uyghurs in the region when detected with the card gel antiglobulin method. Results In 894 specimens,328 Fy (a+b+) cases (36.69%),494 Fy (a+b-) cases (55.26%),and 72 Fy (a-b+) cases (8.05%) were seen but no Fy(a-b-) was observed. The frequencies of Fya and Fyb are 0.7360 and 0.2640,respectively. The distribution of Duffy blood group antigens is in accordance with the Hardy-Weinberg Equilibrium in the Uyghurs. Conclusion The distribution of Duffy blood group antigens of Uygur population in the north region of Xinjiang presents its racial characteristics.

Objective To explore the distribution features of ABO,Rh,MN,and P blood group among RhD negative blood donors in Liuzhou area. Methods 459 RhD negative blood donors were confirmed by indirect antiglobulin test. These blood donors were detected ABO blood group,Rh (C,c,E,e)antigens,MN antigens and P blood group. The gene frequencys were calculated. Results The distribution of ABO blood group:O(0.414 0)>A(0.285 4)>B(0.233 1)>AB(0.067 5); the distribution of gene frequency:r(0.645 2)>p(0.193 4)>q(0.161 4). The distribution of Rh blood group:dccee(0.518 5)>dCcee(0.322 5)>dCCee(0.091 5)> dccEe(0.047 9)> dccEE(0.008 7)=dCcEe(0.008 7)>dCCEe(0.002 2),dCcEE and dCCEE could not be detected; the distribution of gene frequency:cde(0.701 1)>Cde(0.259 6)>cdE(0.039 3)>CdE(0.000 0). The distribution of MN blood group:MN(0.568 6)>M(0.250 6)>N(0.180 8); the distribution of gene frequency:M(0.534 9)>N(0.465 1). The distribution of P blood group:P2(0.684 1)>P1(0.315 9); the distribution of gene frequency:P2(0.827 1)>P1(0.172 9). Conclusion On the whole,the distribution features of ABO,Rh,MN and P blood group among RhD negative blood donors in Liuzhou area are accord with the southern Chinese population.

Objective To investigate the effect of chyle levels of plasma turbidity plate and degrees of plasma hemolysis shadeguide for reducing the unnecessary retirement of plasma due to objective incorrect assessment in the process of blood collection and supply. Methods Fifteen staffs were randomly grouped to compare the chyle and hemolysis samples in critical level at different light by chyle plasma turbidity plate and plasma hemolysis shadeguide in order to check the influence of the natural or artificial light. The chyle and hemolysis plasma discarding data were analyzed from 2012 to 2013. Results There was no difference between the natural light and the light of lamp(P>0.05)whereas a significant difference was seen between chyle levels of plasma turbidity plate and degrees of plasma hemolysis shadeguide(P<0.05). In the year of 2012,the rate of chyle plasma discarding was 0.46%,and that of hemolysis plasma discarding was 0.26%. In 2013,however,the rate of chyle plasma discarding was 0.41%,and that of hemolysis plasma discarding was 0.18%.The rates of both plasma discarding have been obviously reduced after using chyle levels of plasma turbidity plate and degrees of plasma hemolysis shadeguide (P≤0.01). Conclusion The chyle levels of plasma turbidity plate and degrees of plasma hemolysis shadeguide may effectively control the unnecessary retirement of chyle and hemolysis plasma,and can be used as a convenient and effective method to distinguish the chyle and hemolysis plasma in the process of blood collection and supply.

A Comparative Study on the Applicalion of Nucleic Acid Test and ELISA in Blood Donors

Objective To analyze the results of blood virus screening by both nucleic acid test (NAT) and ELISA to evaluate the significance of NAT in improving blood safety. Methods We randomly collected 19 464 blood samples from volunteers between 2015 and 2016. The blood samples were detected by ELISA for screening of anti-HIV,anti-HCV and HBsAg,followed by PCR fluorescence detection for nucleic acids of HBV,HCV and HIV in the samples which had showed negative reactions in ELISA. The comparative results were evaluated for the efficiency and significance in laboratory diagnoses and blood safety ensurance. Result The positive rate of HBV was 16.0% by NAT compared with 11.0% by ELISA (P<0.05). HCV detected by NAT revealed a positivity of 1.8% ,which is obviously higher than 0.5% by ELISA (P<0.05). The positive rate of HIV,however,was 1.9% by NAT whereas 1.7% by ELISA,no significant difference was noted between the two methods (P>0.05). Conclusion Pre-examination of blood samples with ELISA followed by NAT verification is recommended for blood screening which may help avoid missing or misdetection of samples in ELISA and help improve sensitivity and accuracy in the safety of blood transfusion.

A Compararison of ELISA with FQ-PCR for Detection of Anti-HCV Antibodies in the Samples Near Critical Value

Objective To Use the fluorescence quantitative PCR (FQ-PCR) for confirming antibodies against hepatitis C surface antigen(anti-HCV)that had been tested by enzyme linked immunoassay(ELISA). Methods 1 348 patients who had experienced blood transfusion and operation were collected from April 2012 to March 2016.The patients were divided into three groups,group A included 240 samples with negative anti-HCV detected by ELISA (S/CO<0.3); group B covered 902 samples with anti-HCV on cutoff value (0.7≤S/CO<1); and group C included 206 samples with weak reaction of anti-HCV detected by ELISA. Results Among groups B and C,41(4.54%)and 172(83.49%) samples were found to have HCV RNA level>1 000 IU/ml rechecked by FQ-PCR. FQ-PCR demonstrated a high positive rate in the samples on the cutoff value tested by ELISA double reagents(P<0.05); while no difference was noted between the samples with weak reaction of HCV RNA when tested with ELISA and FQ-PCR(P>0.05). Conclusion The false negative and misdiagnosis rate of ELISA is high in the samples near HCV cutoff value and in weak reaction,suggesting that FQ-PCR is useful for early diagnose of HCV patients.

Objective To reveal the potential risk of the negative samples near the critical value in detection of human immunodeficiency virus antibody by chemiluminescence immunoassay. Method The samples were selected by chemiluminescence immunoassay(CLIA) for antibodies against HIV in which absorbance of the sample's cutoff value(S/CO)between 0.3 and 0.9,and checked by enzyme linked immunosorbent assay(ELISA). Result One hundred and thirty-four samples over 0.3<S/CO<0.9 were screened from 64425 clinical samples by CLIA. Among them,2 cases were positive withr ELISA. The two positive cases,however,turned to be negative four weeks later. Conclusion Detection of CLIA HIV antibodies based on the threshold of 0.3<S/CO<0.9 will be able to ensure the safety in HIV diagnosis.

Objective To evaluate the stability of a enzyme-linked Immunosorbent Assays Kit for Anti-HTLV,and to provide references for choosing Anti-HTLV ELISA kits which used in blood screening. Methods Coefficient of variation was used toevaluate intra-assay variation,day-assay variation and inter-assay variation of 4 Lot Numbers. Box-type diagram was used to analyze extreme value. Results The intra-assay variation was 6.5%,none extreme value in all intra-assay results. Coefficients of variation were 6.8%,7.1%,8.8% and 8.6,respectively. Conclusion The reagent has shown reasonably stability in intra-assay,day-assay and inter-assay,which can satisfy the requirement of blood screening kit.

Objective To explore parameter settings of ELISA equipment which is commonly used by blood stations on STAR automatic pipetting instrument,in order to improve pipetting accuracy and efficiency. Methods The instrument pipetting target was set according to the sample volume of each reagent required specification. The differences between actual and target amount of sample were compared with quality pipette instrument,and continuously been amended. Finally a correction was set. Results We explored a STAR pipetting curve of ELISA reagents commonly used by blood station,set a correction for each common target value of ELISA detection of blood station,and optimize the pipetting sequence of 8 kinds of reagents tested and left template. Conclusion Using general parameters provided by factory or specific parameters set by engineer for users can cause some deviations between the target values the actual ones. To ensure accurate and efficient,users are required to set the correction and optimize the pipetting sequence according to the actual pipetting volume in the routine experimentation.
