Application of Nucleic Acid Test in Blood Screening of Donators

Abstract: Objective To investigate the necessity of nucleic acid test (NAT) and its significance in blood screening of donators in Yancheng district. Method sA total of 55 186 blood samples with at least one negative ELISA were selected in 2015. The NATs were performed in all samples based on Roche Cobas s201 nucleic acid amplification testing system (Roche system),the HBV DNA,HCV RNA and HIV RNA were individually detected with DAAN system in the pooled blood of 6 or 8 samples. The positive samples in the screening were subjected to confirmatory tests in the Province Blood Center. Results Totally,7 965 pools were tested by NATs in the samples that showed at least one negative ELISA. Among them,47 (1.20%) positive pools of 3 930 pools were found by Roche system. In the 4 035 pools tested by DAAN system,39 (0.97%) positive pools were noted. 42(0.076% ) samples were found to be positive in individual NATs. The positive rates of Roche system and DAAN system were 0.12% and 0.047%,respectively (P<0.05). In 36 samples,25 (69.44%) were confirmed to be HBV DNA positive,while no HCV RNA and HIV RNA were found. Conclusion Nucleic acid test is recommended following serological detections,which would cover the shortage of serological tests and ensure the blood safety.
